Experiment Title Isolation and screening of photosynthetic microbes as single cell protein source

Experiment Background Crude or refined protein derived from algae, bacteria, mold, or yeast that is used for human or animal consumption is referred to as single-cell protein (SCP). Due to its high protein content, the synthesis and use of microbial biomass as a food source has drawn special attention as an alternative to proteins originating from agriculture. SCP contains lipids, vitamins, and other nutraceutical molecules in addition to proteins. The term "algae as a source of SCP" can refer to either prokaryotic cyanobacteria or small single-cell true algae, both of which grow by using light energy and carbon dioxide (autotrophic growth). Algae are frequently cultivated using methods similar to conventional agriculture, in contrast to other SCP-producing organisms, since depend on large areas and sunlight radiation. Hence, present study aims to isolate and explore their its biochemical characterization for SCP

Treatment
  • Isolation and purification of photosynthetic microbes. For isolation, fresh and marine water samples will be collected.
  • Screening for higher protein production. Spirulina (available in tablet form will be taken from market) will be kept as positive control
  • Estimation of other nutraceuticals compounds (antioxidant activity, total phenol, Flavonoid, Ascorbic Acid)
  • Identification of potential isolate by 16S rDNA/18S rDNA 

Methodology

Experiment will be carried out as described in following flow chart :

  • Samples collection - fresh and marine water
  • Purification of microbes
  • Morphological and growth characteristics of pure culture
  • Screening of isolates for higher protein content
  • Potential microbes will be identified using 16S rDNA/18S rDNA
  • Pure culture will be grown for mass production followed by drying
  • Total phenol, flavanoid and ascorbic acid content will be quantified from dry cell biomass

 

All the parameters will be monitored as per standard reference mentioned below :

  • Protein – Protein estimation will be carried out from dry cell biomass using Folin and Lowery’s method
  • Total phenol – Total phenol will be estimated from dry cell biomass using Slinkard and Singleton, 1977
  • Flavonoid  - Flavanoids from dry cell biomass will be estimated as method described by Zhishen et al., 1999
  • Ascorbic Acid – Ascorbic acid content will be estimated using Loeffler and Ponting, 1942

Identification – Potential protein containing microbes will be identified as method described by Murthy & Vyas, 2015

Observation to recorded :

  • Morphological and growth characteristics of pure culture
  • Protein content (µg/g dry weight)
  • Total phenol (µg/g dry weight)
  • Flavonoid  (µg/g dry weight)
  • Ascorbic Acid (µg/g dry weight)
