Experiment Title Isolation and characterization of chitinolytic bacteria

Experiment Background After cellulose, chitin is the second most abundant natural polymer present on earth. It is found in insects, arthropod, fungi and algae. Many microorganisms produce chitanase which breakdown chitin in its oligo and monomers. Several bacteria reported for chitinolytic activity are Flavobacterium, Bacillus, Cytophaga, Pseudomonas, Aeromonas, Enterobacter, Chromobacterium, Arthrobacter, Serratia, Erwinia, Vibrio etc. Several fungi like Aspergillus, Mucor and Mortierella are also reported for chitinase production. Many fungi and insect contains chitin in their exterior cellular structure. Once this chitin is degraded from insect and fungi, they suffer from severe damage which leads to death. In agriculture field wide variety chemical are used as insecticides and fungicides for disease control. Despite the fact that their ability to control pest and pathogenic fungi, they have severe harmful effect on animal, human and environment. Hence, an alternative solution for control of pathogenic fungi and insect should be developed. Microorganisms which have ability to produce chitinase can be used as biocontrol agents as an ecofriendly approach. Other than agriculture, chitinase has a wide-range of applications in pharmaceutical, food industries, waste management and in control of malaria transmission. Hence, present study aims to isolate and characterize chitinolytic microorganisms.

Treatment

Experiment detail  

  • Isolation and purification of chitinolytic bacteria (by enrichment technique). Samples will be collected from composting and dumping sites 
  • Screening for chitinase production (Qualitative (plate assay) and quantitative assay (Schales' method))
  • Optimization of environmental parameter for potential isolate
  • Identification of potential isolate by 16S rDNA
  • Antagonistic activity against plant pathogens
